Building biomimetic microenvironments with 3D CoSeedis™

Creating a biomimetic microenvironment for cells to grow in is essential for the generation of physiologically relevant 3D spheroids/organoids. Stimulating cells with appropriate clues from the extracellular matrix (ECM) is a key factor to induce proper cell differentiation and essential to the formation of 3D structures that reflect the observed in vivo situation. In addition, growing cells under macromolecular crowding conditions may further induce or increase the deposition of appropriate ECM molecules and consequently help building up a biomimetic microenvironment (reviewed in Benny and Raghunath, 2017; DOI: 10.1177/2041731417730467).
With 3D CoSeedis™, abc biopply provides a flexible and validated platform that integrates the generation of biomimetic microenvironments in an easy and straight forward fashion:
  • Feeder cells can be used in contact with test cells or in distance. When grown in distance, they can be used in 2D monolayers or in 3D structures to build up the appropriate environment for the test cells.
  • Crushed ECM can be seeded into the 3D CoSeedis™ matrix to create a suitable environment for subsequent test cells.
  • ECM components can be added to growth media in order to support test cells with the appropriate clues and factors.
  • High diffusion capability of 3D CoSeedis™ matrix allows the use of crowding macromolecules to induce biomimetic effects during the deposition of ECM by feeder and test cells.
  • The modular 3D CoSeedis™ system allows the easy and reproducible combination all the supporting measurements listed above. Furthermore, such a setup guarantees easy access to individual cells and 3D construct populations for downstream analysis, including histology, in-chip microscopy, profiling of individual spheroids/organoids, and many more.
